WB-STIM — Whole Blood Stimulation Buffer
In short
WB-STIM lets you run a stimulation assay directly on whole blood — no PBMC isolation, no granulocyte loss, no operator-introduced variability.
Mix it 1:1 with fresh blood, add your stimulant, and the immune response you measure afterward is closer to what's actually happening in the body.

How it fits your workflow
Standard sequence: fresh whole blood → WB-STIM (1:1) → stimulate → then either:
- Freeze in Stabiliser (1:1 with the full stimulated suspension) for later processing — recalculate fixation buffer at ×11.5 instead of the standard ×10 when you get to the Fix-Lyse step (Protocol P01/P07); or
- Fix immediately for short time-course work needing second-to-minute resolution, since fixation locks in the proteome within seconds — then freeze the fixed sample in CRYO#20 or stain straight away.
One rule has no exceptions: stimulate before you stabilise. Blood already preserved in the Stabiliser cannot be used for a stimulation assay afterward — see Protocol P07.
Technical specifications
- Serum-free, endotoxin-free. Standard formulation contains antibiotics and phenol red; an antibiotic-/phenol-red-free version is available on request (more physiologically compatible with whole blood, but requires stricter GLP handling — the trade-off is a lower risk of masking a pre-existing infection in the reagent).
- Blood-to-WB-STIM ratio: 1:1.
- Validated stimulants — LPS, R848, Poly I:C, IFNβ — are stable for at least 3 months at −20°C when frozen separately from the WB-STIM buffer, then combined after thawing.
- Adding a custom stimulant, activating antibody, or tumour cell line hasn't been directly tested, but no obvious limitation is expected.
- Optimized and compatible with: Whole Blood Cell Stabiliser, Fix-Lyse Kit, CRYO#20, and Protocols P01/P03/P06/P07.
