How long can blood wait before I add the Stabiliser?
Up to 24 hours works for most donors. Mix within 30 minutes to 2 hours when you can. A gentle tube rocker helps during the wait.
What anticoagulant should I use — heparin or EDTA?
Both work. Heparin is our default recommendation — the leftover blood stays usable for stimulation assays, which EDTA-treated blood is not.
How much blood do I actually need?
As little as 50–100 µl for standard immunophenotyping. Plan for 200–500 µl if CyTOF or deep flow panels are the endpoint.
Do I need a controlled-rate freezer (e.g. Mr. Frosty)?
No. Tubes go straight into a -80°C freezer.
What's the shelf life and storage temperature?
24 months minimum, refrigerated at 2–8°C. Short excursions to room temperature (days, even a couple of weeks) don't affect quality.
What does each buffer in the kit do?
Fix-Concentrate and Fix-Diluent combine 1:1 to make the Fixation buffer. LYSIS removes red blood cells — but only when used together with the Fixation buffer, never alone. WASH stabilises the pellet and clears lysis remnants.
Do I need a separate permeabilisation buffer for intracellular staining?
